Denaturing gradient gel electrophoresis (DGGE) and SYBR Green I quantitative real-time PCR (qPCR) approaches were used to assess respectively the molecular diversity and the quantity of the nifH gene sequences in the rhizospheres of two cultivars of sorghum sown in Cerrado soil with contrasting levels of nitrogen fertilizer. DGGE fingerprinting showed that for both cultivars the presumptive nitrogen-fixing populations in the rhizospheres were more diverse than in bulk soil. Sequencing of nifH gene fragments obtained from DGGE bands revealed that members of the order Rhizobiales were prevalent among the dominant diazotrophs. Discriminant analysis of DGGE profiles resulted into three groups formed by (i) cultivar BRS 308 sown with high level of nitrogen, (ii) cultivar BRS 308 sown with low level of nitrogen and cultivar BRS 310 sown either with low or high levels of nitrogen and (iii) bulk soil, showing that the nitrogen fertilization influenced the nifH gene sequence diversity only in the rhizosphere of cultivar BRS 308. When the quantity of the nifH gene sequences was determined by q-PCR, 2.4 x105 to 1.3 x 107 copies/g of soil were detected. The highest number of nifH gene copies was observed in the rhizosphere of cultivar BRS 308 treated with low amount of fertilizer, and a reduction in nifH density was observed in the rhizospheres of both sorghum cultivars when high levels of nitrogen were applied. Thus, both the amount of nitrogen fertilizer and the cultivar are important factors influencing the structure and amount of diazotrophs in sorghum. Muitos micro-organismos endofíticos têm sido descritos por apresentarem promissores potenciais de aplicação biotecnológica, incluindo a capacidade de promoverem o crescimento de plantas, como o milho. Este trabalho teve como objetivos isolar bactérias endofíticas associadas às folhas, seiva e raízes do cultivar híbrido de milho Pionner 30F35 Herculex, cultivado com e sem adubação fosfatada, e analisar sua capacidade de produzir fatores promotores de crescimento vegetal. Um total de 178 bactérias foram isoladas e identificadas pelo sequenciamento parcial do gene rDNA 16S. Cerca de 70% dos isolados foram classificados como gram-positivos, sendo que a proporção deste grupo foi maior entre os isolados de seiva (82%), seguido por folhas (76%) e raízes (57%). Houve uma distribuição equivalente de isolados pertencentes aos clados Actinobacteria, Firmicutes e Proteobacteria, e apenas um isolado (Flavobacterium acidificum RT3B-41) do filo Bacteroidetes foi encontrado. Os principais gêneros encontrados foram Microbacterium, Bacillus, Staphylococcus, Pseudomonas, Lactococcus, Enterobactere Curtobacterium. In vitro, 81% dos isolados presentaram pelo menos um atributo potencialmente promotor de crescimento vegetal, sendo que 57% dos isolados apresentaram a capacidade de produzir moléculas do tipo auxina, 31% foram capazes de solubilizar fosfato inorgânico, 42% de mineralizar fitato, 15% de antagonizar bactérias e fungos fitopatogênicos, e 43% dos isolados de seiva e folhas foram capazes de produzir sideróforos. As densidades bacterianas também foram computadas e variaram com a parte da planta analisada (folha, seiva e raiz), o que não aconteceu em relação aos tratamentos de adubação fosfatada. Porém, tanto as partes das plantas quanto os tratamentos de adubação apresentaram efeito significativo sobre a frequência de isolados produtores de fatores de crescimento vegetal, assim como sobre suas taxas de produção ou atividade. Estes resultados permitirão em trabalhos futuros considerar o uso destes isolados como bioinoculantes promotores de crescimento no momento de plantio do milho e como agentes de controle biológico, visando o aumento de produtividade desta cultura.
